چکیده

Fear extinction requires coordinated neural activity within the amygdala and medial prefrontal cortex (mPFC). Any behavior has a transcriptomic signature that is modified by environmental experiences, specific genes are involved in functional plasticity synaptic wiring during fear extinction. Here, we investigated effects of optogenetic manipulations prelimbic (PrL) pyramidal neurons gene expression to analyze transcriptional pathways associated adaptive maladaptive To this aim, transgenic mice were (or not) fear-conditioned phase they received sham) stimulations over photo-activable PrL neurons. At end behavioral testing, electrophysiological (neural cellular excitability Excitatory Post-Synaptic Currents) morphological (spinogenesis) correlates evaluated Furthermore, cell-specific RNA-analyses (differential profiling enrichment analyses) performed Our results show activation induces deficits, reflected an increase excitability, excitatory neurotransmission, spinogenesis neurons, strong modifications transcriptome Understanding electrophysiological, morphological, architecture may facilitate comprehension fear-related disorders.

Optogenetic drive of neocortical pyramidal neurons

Local fluctuations in the blood oxygenation level-dependent (BOLD) signal serve as the basis of functional magnetic resonance imaging (fMRI). Understanding the correlation between distinct aspects of neural activity and the BOLD response is fundamental to the interpretation of this widely used mapping signal. Analysis of this question requires the ability to precisely manipulate the activity of...

Specific Targeting of the Basolateral Amygdala to Projectionally Defined Pyramidal Neurons in Prelimbic and Infralimbic Cortex.

Adjacent prelimbic (PL) and infralimbic (IL) regions in the medial prefrontal cortex have distinct roles in emotional learning. A complete mechanistic understanding underlying this dichotomy remains unclear. Here we explored targeting of specific PL and IL neurons by the basolateral amygdala (BLA), a limbic structure pivotal in pain and fear processing. In mice, we used retrograde labeling, bra...

Optogenetic stimulation of mPFC pyramidal neurons as a conditioned stimulus supports associative learning in rats

It is generally accepted that the associative learning occurs when a behaviorally neutral conditioned stimulus (CS) is paired with an aversive unconditioned stimulus (US) in close temporal proximity. Eyeblink conditioning (EBC) is a simple form of associative learning for motor responses. Specific activation of a population of cells may be an effective and sufficient CS for establishing EBC. Ho...

Pyramidal Neurons

The effect of the fluorescent Ca2+ indicator dye Fura-2 on Ca2+ dynamics was studied in proximal apical dendrites of neocortical layer V and hippocampal CAl pyramidal neurons in rat brain slices using somatic whole-cell recording and a charge-coupled device camera. A single action potential evoked a transient increase of intradendritic calcium concentration ([Ca2+]i) that was reduced in size an...
